Abstract
BACKGROUND: Nicolaides-Baraitser syndrome (NCBRS) is a rare disorder characterized by neurodevelopmental delays, seizures, and diverse physical characteristics. The DNA methylation (DNAm) profile in NCBRS is significantly different. DNAm is linked to the biological aging of cells and the health risks associated with biological aging. In this study, we examined changes in biological ages in NCBRS to provide insights into the prognosis and health risks of NCBRS.Entities:

FIGURE 1PhenoAge, GrimAge, and DNAmTL of the GSE125367 dataset. (a–c) Scatter plots were created using PhenoAge/GrimAge/DNAmTL versus chronological age. The correlations of PhenoAge/GrimAge/DNAmTL with chronological age were studied by calculating the Pearson's correlation coefficient. PhenoAge and GrimAge showed a significant positive correlation with chronological age in the NCBRS and control groups. DNAmTL showed a significant negative correlation in the control group and a trend of negative correlation in the NCBRS group. (d–f) Violin plots with dots show the acceleration of PhenoAge (AgeAccelPheno) and GrimAge (AgeAccelGrim), as well as the age‐adjusted estimate of DNAmTL (DNAmTLadjAge) in NCBRS cases and controls. p‐values are shown the results of multiple linear regression analysis
