Abstract
BACKGROUND: Promoter hyper-methylation of tumor suppressor genes is a common event that occurs in cancer. As methylation is a reversible modification, agents capable of reversing an abnormal methylation status should help to combat cancer. 5-Aza-CdR is a DNA methyl-transferase inhibitor. The present study aimed to evaluate the effect of 5-Aza-CdR on the proliferation of human pancreatic cancer cell line (PANC-1) and the expression of RASSF1A and Bax genes.Entities:

Figure1The morphologic view of PANC-1 after treatment with 5 and 10 µm of 5-Aza-CdR (×100). Clearly, the percentage of survived cells has significantly decreased after 96 hours of treatment with 10 µm of 5-Aza-CdR.
Figure25-Aza-CdR treatment in PANC-1 (5 and 10 µM for 96 h). 10 µm of 5-Aza-CdR epigenetically induced re-expression of RASSF1A (P=0.042) and subsequently up-regulation of Bax genes (P=0.034). However, in a concentration of 5 µm, the expression ratio of these genes was not significantly different in comparison with controls (P=0.784).
