Figure 1(a) Recombinant AtPLDα activity in crude protein extracts of Pichia pastoris transformed with the empty vector or expressing either the recombinant AtPLDα-WT or the AtPLDα-6xHis, after 4 days of culture, and using POPC (0.26 mM, final concentration) as the substrate. Values presented are the means ± SD obtained from four independent PCR-positive clones. P < 0.01 (versus blank). (b) SDS-PAGE (10% acrylamide) analysis of the recombinant AtPLDα-WT purified using the purification procedure with octyl-Sepharose CL-4B column (2.5 × 20 cm) (see Materials and Methods). Lane 1: molecular mass marker, lane 2: eluted fraction containing the recombinant AtPLDα-WT. The gel was stained with Coomassie Brilliant Blue R-250 to reveal the proteins. The arrow on the right indicates the position of the AtPLDα-WT.
Figure 2Protein sequence alignment of the N-terminal region of purified α-type PLDs reported so far. Grey-colored residues represent predicted sequences deduced from cDNAs and absents in mature PLD forms (black-colored residues). Residues highlighted in yellow come from the Edman degradation sequencing obtained in this study while the posttranslational acetylated residue is highlighted in green.
Figure 3(a) SDS-PAGE analysis of crude protein extracts from E. coli cells expressing either the C2α domain or the C2β domain, in comparison to the empty vector as expression control. Lane 1: molecular mass marker, lane 2: empty vector, lane 3: C2α domain, and lane 4: C2β domain. (b) SDS-PAGE analysis of affinity purified C2α domain and C2β domain. Lane 1: molecular mass marker, lane 2: C2α domain, and lane 3: C2β domain. Protein migrations were performed using 15% SDS-PAGE and stained with Coomassie Brilliant Blue R-250.
Figure 4Intrinsic fluorescence spectra (λ ex 280 nm) of urea-denatured C2α domain (a) and C2β domain (b) diluted in a Tris/HCl buffer containing (red curves) or not (blue curves) 8 M urea and after 1 h of incubation at room temperature. The final concentration of both domains was set at 1 μM, leading to a final concentration of urea ranging from 50 mM to 75 mM for blue curves. Fluorescence intensity was expressed in arbitrary units (AU) and bandwidths were set at 10 nm for both excitation and emission.
Figure 5(a) Lipid blot analysis of the interaction of the C2α (left panel) and the C2β (middle panel) in the presence of 1 mM of Ca2+ or 1 mM of EDTA, in comparison to the dialysis buffer serving as the negative control (right panel). Lipids were spotted onto a PVDF membrane and interactions were revealed using an anti-6xHis antibody, as described in the experimental procedures. TG: triglyceride, +: positive control made by spotting 6xHis-C2 domain protein. (b) Relative binding of the C2α (left panel) and the C2β (right panel) to phospholipids vesicles, using the protein to dansyl-PE FRET. Blue bars: no Ca2+ added, red bars: 50 μM of Ca2+, and green bars: 200 μM of Ca2+. Values presented are the means ± SD obtained from three independent experiments.
Figure 6(a) Multiple sequence alignment of C2 domains. Amino acid sequences of C2α and C2β were aligned with various known C2 domain structures (see Materials and Methods). Residues highlighted in red represent Ca2+ binding residues extracted from structures and residues highlighted in green represent membrane binding residues extracted from the OPM database. The proposed α-helix between β1 and β2 strands is indicated in blue. CBR: Ca2+ binding region. Strand β0, colored in yellow, corresponds to the first β strand present in typology I. The observed cleavage on the recombinant AtPLDα is indicated by the arrow (red). The PDB entries correspond to cPLA2 (1RLW), perforin (3W57), Munc13-1 (3KWU), Car4 (5A50 and 4V29), Car4 (5A52), PKCα (1DSY), PKCβ (1A25), rabphilin-3A (2K3H and 2CM5), Synaptotagmin-1 C2A (1BYN), and C2B (1UOV), SV2A (4V11), and Synaptotagmin-7 C2B (3N5A). Modeled structures of the C2α domain (b) and the C2β domain (c), using the I-tasser server and docked to membrane (dummy) using the PPM server. Ca2+ is colored in pink.
