Abstract
BACKGROUND: Pathogenic Leptospira spp. present in the blood of patients with leptospirosis during the first week of symptoms can be detected using culture or PCR. A proportion of patients who are positive by PCR are negative by culture. Leptospira spp. are fastidious bacteria, and we hypothesized that a false-negative culture result may represent infection with a distinct bacterial subset that fail to grow in standard culture medium.Entities:

Figure 1Phylogenetic analyses of 184 partial . The tree was reconstructed using the neighbor-joining method as implemented in MEGA ver 5.0 [15]. A total of 45 polymorphic sites were demonstrated. The vertical numbers shown indicate the nucleotide position based on rrs of L. interrogans serovar Lai [GenBank: AE01300.1], and a dot indicates an identical nucleotide at that position.

Figure 2Phylogenetic analysis of a 443-nucleotide region of . The tree was reconstructed using the neighbor-joining method as implemented in MEGA ver 5.0 [15]. Colour coding represents the following: RED, sequence of a PCR fragment amplified from patients with culture-positive leptospirosis; BLUE, sequence of a PCR fragment amplified from patients with culture-negative leptospirosis; YELLOW, sequence for reference strains obtained from GenBank.
Figure 3Comparison of polymorphic sites in a 443-nucleotide region of . The sequence of L498 was compared with that of four species (nine different rrs alleles) with which it genetically clusters.
