Determination of protein absorption coefficients using a refractive index chromatographic monitor.

Abstract

Using lysozyme as a primary standard, a refractive index monitor designed for column chromatography was used to determine protein concentration and hence absorption coefficients. The method is non-destructive, requires only small amounts of protein approximately 0.2 mg, and could be adapted for smaller samples.
