Abstract
INTRODUCTION: Isoprenoids are amongst the most abundant and diverse biological molecules and are involved in a broad range of biological functions. Functional understanding of their biosynthesis is thus key in many fundamental and applicative fields, including systems biology, medicine and biotechnology. However, available methods do not yet allow accurate quantification and tracing of stable isotopes incorporation for all the isoprenoids precursors.Entities:

Cells are quenched in liquid nitrogen after fast filtration, and intracellular metabolites are extracted by incubating the filters during 10 min in 5 mL of isopropanol/H2O 100 mM NH4HCO3 (1:1) at 70 °C. The extract is centrifuged, dried, resuspended in 200 µL of methanol/NH4OH 10 mM (7:3) at pH 9.5, and analyzed by LC–HRMS.
