随着多药联合化疗的应用,急性淋巴细胞白血病(ALL)的治愈率得到了极大的提高,但仍有15%~20%的ALL患儿难以到达长期缓解[1]。白血病细胞耐药是导致白血病治疗失败的最主要原因[2]–[3]。因此寻找区别于目前临床治疗药物的新类型化疗靶向药物,有可能克服常用药物形成耐药的问题。近年来研究发现的一种天然小分子化合物—Rita(Reactivation of p53 and induction of tumor cell apoptosis)能激活细胞内P53蛋白,促进肿瘤细胞凋亡,可能成为肿瘤治疗的新药物[4]。目前关于Rita的报道多集中在实体瘤[5]–[7],ALL中鲜有报道。尽管ALL中P53表达阳性的比例仅占10%[8],但54%复发耐药的ALL患者表达P53[9]。Rita能否通过恢复耐药ALL细胞中P53的功能,从而改善耐药ALL的治疗效果,目前尚不明确。我们提供探讨Rita在P53野生型ALL细胞系中的功能及调控机制,以期为ALL治疗引入新型化疗靶向药物提供理论依据。
