Abstract
PURPOSE: Limited studies have shown antifibrotic effects of pentoxifylline, captopril, simvastatin, and tamoxifen. No comparisons are available of the effects of these drugs on prevention of renal and bladder changes in partial urethral obstruction (PUO).Entities:

The weight and volumes in the sham-operated and PUO groups
Values are presented as mean±standard deviation.
PUO, partial urethral obstruction; PCT, proximal convoluted tubule; DCT, distal convoluted tubule; CD, collecting ducts; LH, loop of Henle; IT, interstitial tissue; NS, normal saline; TAM, tamoxifen; CAP, captopril; PEN, pentoxifylline; SIM, simvastatin.
*p<0.05, PUO+NS vs. (PUO+TAM), (PUO+CAP), (PUO+PEN), and (PUO+SIM). **p<0.01, Sham-operated vs. PUO+NS. †p<0.05, PUO+PEN vs. (PUO+TAM), (PUO+CAP), and (PUO+SIM).
Length (m) of the PCT, DCT, CD, LH, and vessel in the sham-operated animals and the rats treated with NS, TAM, CAP, PEN, or SIM after PUO
Values are presented as mean±standard deviation.
PCT, proximal convoluted tubule; DCT, distal convoluted tubule; CD, collecting ducts; LH, loop of Henle; NS, normal saline; TAM, tamoxifen; CAP, captopril; PEN, pentoxifylline; SIM, simvastatin; PUO, partial urethral obstruction.
*p<0.05, PUO±NS vs. (PUO+TAM), (PUO+CAP), (PUO+PEN), and (PUO+SIM). **p<0.01, Sham-operated group vs. PUO+NS. †p<0.05, PUO+PEN vs. (PUO+TAM), (PUO+CAP), and (PUO+SIM).
Weight of the bladder (mg), volume (mm3) of the bladder, epithelial, interstitial, and muscular tissues, and the total number of the fibroblast and fibrocytes in the sham-operated animals and the rats treated with NS, TAM, CAP, PEN, or SIM after PUO
Values are presented as mean±standard deviation.
NS, normal saline; TAM, tamoxifen; CAP, captopril; PEN, pentoxifylline; SIM, simvastatin; PUO, partial urethral obstruction.
*p<0.05, PUO+NS vs. (PUO+CAP), (PUO+PEN), and (PUO+SIM). **p<0.01, PUO+NS vs. Sham-operated. †p<0.05, PUO+PEN vs. (PUO+TAM), (PUO+CAP), and (PUO+SIM). ‡p<0.05, PUO+NS vs. (PUO+PEN), and (PUO+SIM).
FIG. 1Micrograph of the kidney in the different groups. (A) Sham-operated animals with normal structure. (B) PUO+normal saline treatment. The normal tubular cells are lost. Fibrous tissue was formed and replaced the tubular tissue. The tubules seemed dilated. (C) PUO+PEN, (D) PUO+CAP, (E) PUO+SIM, (F) PUO+TAM. Heidenhain's AZAN trichrome stain (scale bar, 50 µm). PUO, partial urethral obstruction; PEN, pentoxifylline; CAP, captopril; SIM, simvastatin; TAM, tamoxifen.
FIG. 2Micrograph of the bladder in the different groups. (A) Sham-operated animals with normal structure. (B) PUO+ NS, (C) PUO+PEN, (D) PUO+CAP, (E) PUO+SIM, (F) PUO+TAM. PUO without pharmacological treatment showed that more epithelial tissue can be seen. The muscle layer was hypertrophied. Fibrous tissue was increased in the different layers of the bladder. PUO+PEN ameliorated the changes in bladder structure followed by CAP, SIM, and TAM. Heidenhain's AZAN trichrome stain (scale bar, 250 µm). PUO, partial urethral obstruction; PEN, pentoxifylline; CAP, captopril; SIM, simvastatin; TAM, tamoxifen.
