Abstract
Dynamic (-) DNA supercoiling generated in the wake of translocating protein complexes is known to occur during transcription. Recent studies indicate that (-) superhelical tension also builds up specifically in the leading duplex during replication. Here, we argue that this unrecognized supercoiling is causally involved in the regulation of key DNA transactions and deserves further consideration.Keywords: DNA polymerase; DNA transactions; dynamic DNA supercoiling; leading strand duplex; parental DNA; protein translocation; replication
