A method for determining the three-dimensional form of active flagella, using two-colour darkground illumination.

Using darkground illumination, and placing colour filters over half the field, one can make a single, highly resolved, colour micrograph of a filamentous object (such as a mammalian sperm tail) from which it is possible to get a complete and unambiguous three-dimensional reconstruction.
