Abstract
Reprogramming of somatic nuclei toward the embryonic state has been studied using nuclear transfer (NT) to an oocyte at the metaphase II (MII) stage. In this NT, a somatic nucleus transplanted into an MII oocyte of the same species undergoes DNA replication and cell division before activating embryonic genes. Here, we describe a direct NT protocol using 4-cell stage mouse embryos that enables reprogramming of intra- and cross-species nuclei to express embryonic genes without requiring DNA replication and cell division. Accumulation of LacR-mCherry onto the transgene site of donor nuclei marks the injected nuclei after NT-ETR (red dots; white arrows). The comparison between 2 h and 24 h after NT-ETR clearly shows the swelling of donor nuclei in the arrested mouse 4-cell embryos, finally resembling the morphology of embryonic nuclei. Figure 3The whole process of NT-ETR
IVF-derived BDF1 mouse embryos are injected with or without LacR-mCherry mRNA solution at 6 hpi and cultured in KSOMaa medium containing 0.1 μg/mL demecolcine from 40 hpi. At 46 hpi, NT of differentiated donor cells is performed. The NT embryos are further cultured for 2 or 24 h and subjected to immunofluorescence staining, qRT-PCR or RNA-seq analyses. IVF represents in vitro fertilization. Figure 3 is modified from the original Figure 3 of Tomikawa et al. (2021).
