Abstract
Purpose: Omentin-1/intelectin (ITLN)1 is an adipocytokine with both anti-inflammatory and anti-oxidative stress properties, and little is known about its role in male reproduction. This study was aimed at exploring the relationships among omentin-1/ITLN1, semen parameters and F2-isoprostanes (F2-IsoPs), a maker of oxidative stress, in groups of patients affected by different pathologies. In addition, omentin-1/ITLN1 immunolocalization was assessed in ejaculated spermatozoa and in tissues of male reproductive system. Patients andEntities:

Figure 2UV micrographs of human sperm of fertile (A and B) and infertile men with varicocele and infection (C and D) treated with an antibody anti- omentin-1/ITLN1. In sperm of fertile men (A and B), the labelling is evident in a spot located in the connecting piece and in the midpiece. In sperm of patients with varicocele (C) and infection (D) the entire tail is labelled. The nuclei are stained with DAPI. Bars: 6 μm.
Figure 3Median (IQR) of percentages of sperm showing different omentin-1/ITLN1 labelling after immunofluorescence. The experiments are performed in samples of fertile men (No. 10) and infertile patients with varicocele and infection (No. 10). * p<0.002; ** p<0.000.
Figure 4Immunolocalization of omentin-1/ITLN1 in epididymis and seminal vesicles (Ultraview Universal Alkaline Phosphatase Red Detection Kit, Ventana). In epididymis (A and B) the immunostaining is present in the basal area of epididymal tubule. Seminal vesicle epithelium (C–F) shows a diffuse signal (C and D) located in the cytoplasm of columnar cells (E). (F) shows a double immunostaining with PAX8 (nuclear marker of seminal vesicle epithelium) located in the nuclei (brown) and omentin-1/ITLN1 located in the cytoplasm (pink/red). Bars: A, B: 50 µm; C, D:100 µm; E: 10µm; F: 6 µm.
