Abstract
Cholinergic neurons control numerous primate-specific and sexually dimorphic brain functions. Here, we present our differentiation protocol for the closely related human female and male neuroblastoma-originated cell lines LA-N-2 and LA-N-5. Pro-cholinergic differentiation (with upregulation of choline acetyltransferase) of both lines can be achieved using neurokines such as ciliary neurotrophic factor (CNTF). Comparative RNA sequencing and mass spectrometry analyses between those two cell lines, supported by experimental intervention, will deepen our understanding of cholinergic systems in human psychiatric and neurologic disease. For complete details on the use and execution of this protocol, please refer to Lobentanzer et al. (2019).Entities:

Figure 1LA-N Differentiation
(A) Timeline of cell seeding, intervention, and lysis/RNA extraction.
(B) Time-dose curves of differentiation of LA-N-2 and LA-N-5. CNTF added to culture medium leads to upregulation of CHAT in a dose-dependent manner (measured relative to ACTB expression). LA-N-2 cells were treated with 1, 10, or 100 ng/mL CNTF; LA-N-5 cells were treated with 1, 2, 10, or 100 ng/mL CNTF. LA-N-5 show an inverse U type dose response, which may also be the case in LA-N-2 when further increasing the dose above 100 ng/mL. ∗p < 0.05, ∗∗p < 0.001. The figure reuses parts of a figure from Lobentanzer et al. (2019).
