Embryonic day 15.5 (E15.5) neurons were dissected from the cortex of mice with indicated genotypes, and cultured for 56 hours prior to addition of proteasomal and deubiquitinase inhibitors for 4 hrs. Half of the plated neurons were treated with 600ng/ml of netrin-1 for 40 min prior to lysis. Neurons were lysed and trypsin digested. Ubiquitylated peptides were enriched and analyzed by mass spectrometry.
