Abstract
Canine kobuvirus (CaKoV) is a causative agent of gastroenteritis in dogs. Rapid detection of CaKoV is important for preventing and controlling this condition. In this study, an SYBR Green I-based quantitative real-time PCR assay was established for CaKoV detection. Specific primers targeting a highly conserved region of the CaKoV 3D gene were developed. After optimization, the method detected a minimum of 1 × 101 copies/μL with high specificity, stability, and repeatability. Moreover, the entire process only required approximately 1.5 h for completion. Our results were supported by those obtained for clinical samples, in which our developed method was successfully applied. The newly established real-time PCR is a rapid, sensitive, specific, and repeatable method for the quantitative detection of CaKoV and can, therefore, be used in epidemiological studies.Entities:

Fig. 1a) Amplification curve of CaKoV by SYBR Green I-based real-time PCR. Ten-fold dilutions of standard template ranged from 1 × 108 to 1 × 101 copies/μL. b) Standard curve of CaKoV by SYBR Green I-based real-time PCR using a standard template diluted 1 × 108 to 1 × 101 copies/μL. The equation for the standard curve for CaKoV was y = -3.359 x + 41.789, with R2 values of 0.999. c) Melting curve. Dilutions ranged from 1 × 108 to 1 × 101 copies/μL, and the Tm value was 80 °C ± 0.5 °C (For interpretation of the references to colour in this figure legend, the reader is referred to the web version of this article).
Fig. 2Sensitivity analysis. The detection limit of the real-time PCR assay was 1 × 101 copies/μL. In contrast, conventional PCR had a detection limit of 1 × 103 copies/μL. The conventional PCR product was about 152bp. Template amounts for curves were 1 × 106–1 × 101 copies/μL and templates for lanes 1–6 were 1 × 106–1 × 101 copies/μL, and lanes 7 was negative control.
Fig. 3Analysis of specificity. Sample 1 represents CaKoV virus, samples 2–6 represent CCV, CPV, CCoV, CDV, and CaAstV, and sample 7 is the negative control.
