Abstract
OBJECTIVES: One percent toluidine blue being the most effective adjunct is often used to detect dysplasia. Not much attention has been given to the effect of toluidine blue in enhancement of cytological smears. The present study assessed the smears before and after application of toluidine blue in smokers against non-smokers using three different stains [Papanicolaou (PAP), Hematoxylin and Eosin (H and E), and Giemsa]. Figure 1Photomicrograph showing exfoliated buccal cells before and after application of toluidine blue (a-b : Papanicolaou stain, x400; c-d: H and E, x400; e-f : Giemsa stain, x400). The pictures show better cytoplasmic detail and background from before to after application of toluidine blue
Figure 2Photomicrograph exhibiting cytological alterations seen in cells of smokers showing (i) Before toluidine blue application (a) clumping/agglomerations of squamous cells (H and E, ×400) (b) bacterial colony unit in squamous cell (Papanicolaou stain, ×400) (arrow) (ii) After toluidine blue application (c) micronuclei (H and E, ×400) (arrow) (d) keratin squame (H and E, ×400) (arrow) (e) cellular pleomorphism of buccal cells (H and E, ×400) (arrow) (f) binucleation of squamous cell (H and E, ×400)
Comparison of smokers and non-smokers with respect to different variables as a whole by Mann-Whitney U test
Comparison of means of all parameters of smokers against non-smokers using all the stains both before and after application of toluidine blue
Comparison of means of all parameters before and after application of toluidine blue in each of the stains in smokers and non-smokers
