CpG Sites that yielded meta-analysis in models before and after heterogeneity adjustments.
Figure 2.Differences in average DMA methylation (DNAM) by cadmium (Cd) as a dichotomous, continuous, and log-transformed variable (panels A, B, and C, respectively), at the two false discovery rate (FDR)-significant CpGs (left, cg11701084; right, cg15345741). Robust linear regression models were adjusted for fetal sex, maternal smoking during pregnancy, mother’s highest achieved education level, putative placental cell types, and an indicator for study [New Hampshire Birth Cohort Study (NHBCS) and Rhode Island Child Health Study (RICHS)]. For models of dichotomous Cd, values were classified as low or high according to the median value for each cohort (3.13 and for NHBCS and RICHS, respectively.).
Figure 3.Manhattan plot of Cd meta-analysis results surrounding the GAS7 gene. Annotated with Ensembl gene names and RefSeq IDs for GAS7 variants, Spearman correlations between GAS7 CpG sites, and CpG-expression correlations. Cadmium (Cd) associations at CpGs outside of the GAS7 coding region () have triangles, whereas CpGs within GAS7 have squares; hits with meta-analysis are indicated with an asterisk (*). Directions of association between DNA methylation (DNAM) and GAS7 expression are indicated with and signs for those relationships whose Spearman correlation p-values were nominally significant ().
