Abstract
The metC gene of Salmonella typhimurium was cloned into the plasmid vectors pACYC184 and pBR322. Genetic and biochemical experiments indicate that the region controlling metC gene expression is present on the cloned fragments. The location of the metC gene was determined by insertional inactivation with transposons Tn5 and mini-Mu. The gene product was identified in a minicell system as a 49-kDa polypeptide. The direction of transcription and translation was determined by correlating the orientation of mini-Mu insertions within the metC gene with the expression of the lacZ gene contained in mini-Mu.Entities:
