Abstract
OBJECTIVE: Thrombin has multiple proatherogenic effects including platelet activation and the induction of inflammatory processes. Recently, the cytokine oncostatin M has been shown to have proinflammatory effects. This study was designed to investigate the effects of thrombin inhibition on the initiation and progression of atherosclerosis and on the expression of oncostatin M.Entities:

Figure 1Effects of dabigatran etexilate on atherosclerotic lesion size within the aortic sinus.
Notes: (A) Treatment with dabigatran etexilate resulted in a significant reduction of the mean lesion area in 9-week-old ApoE–/– mice (P=0.004). (B) administration of dabigatran etexilate also reduced the mean plaque area in ApoE–/– mice at 28 weeks of age (after 20 weeks on a dabigatran etexilate-supplemented diet) (P=0.014).
Figure 2Effects of dabigatran etexilate on atherosclerotic lesion composition in the aortic sinus following 20 weeks of treatment with dabigatran etexilate.
Notes: Average size of the necrotic core (A) in mice at 28 weeks of age (data presented as the ratio of the necrotic core area to the total lesion area) (P=0.02) and average thickness of the fibrous cap (B) (P=0.06).
Figure 3Effects of dabigatran etexilate on the lesion content of oncostatin M (OSM) in the aortic sinus.
Notes: (A) OSM expression within the atherosclerotic lesions of ApoE–/– mice at 28 weeks of age (20 weeks on a drug-supplemented diet) (P=0.04). (B) OSM expression within the atherosclerotic lesions of ApoE–/– mice at 40 weeks of age (20 weeks on dabigatran etexilate) (P=0.01). Data in (A) and (B) are presented as the average area of positive staining for OSM/total plaque area.
