The restriction enzyme target approach to genotyping by sequencing (GBS).

Abstract

The modified genotyping by sequencing method described here emphasizes verifying the success of each library ligation by performing individual PCRs, before preparing the pool of barcoded amplicons to be sequenced. Although this extra step might seem excessive, it will give peace of mind to the researcher knowing each individual is represented in the data set and avoid additional data imputation at the analysis stage.
