Abstract
The gene for cytochromeb-559, associated with the photosystem II reaction center, has been located on the spinach plastid chromosome by cell-free coupled transcription-translation and RNA-programmed hybrid selection translation using appropriate recombinant DNAs, RNA fractions, and monospecific antisera. The gene is located in the large single-copy segment of the plastid chromosome between the genes for cytochomef and the P680 chlorophylla apoprotein of photosystem II and transcribed in the opposite direction relative to these genes. The 10 kd protein is decoded from a bicistronic 1.0 kb mRNA and is apparently not made as a precursor in cell-free rabbit reticulocyte andE. coli lysates.Entities:
