Figure 1Initial chromatographic separation of LNCap cell acid soluble thiol reactants with a three-fold excess of ESSE on a 30 × 2.5 cm column of ODS-AQ gel using 10–15 mL of sample saturated with NaCl. Elution: flow rate 1 mL/min, 200 mL water followed by a 400 mL linear gradient to 40% methanol; 10 mL fractions. (A) 325 nm absorbency trace from a bulk preparation; (B) 35S trace from cells labeled in monolayer (circa 80% confluent) for 4 hours.
Figure 2Anion exchange chromatography of RSSE isolated from LNCap ASF on ODS-AQ (peak 2) using a 30 × 2 cm column of DE52 cellulose. Elution consisted of a 125 mL wash with 20 mM Tris-HCl pH 7.2 containing 5% v/v methanol followed by an 800 mL linear gradient to 0.35 M guanidinium chloride 20 mM Tris-HCl pH 7.2 containing 5% v/v methanol at a flow rate of 1 mL/min. 10 mL fractions were collected. Upper trace—Applied 60 A325 units of RSSE: abscissa A325 × 103; Lower trace—15 A325 units/3.5 × 107 dpm R35SSE: abscissa 35S dpm × 103.
