Oligonucleotide fingerprinting of Hela cell 5 S RNA labeled in vitro with (125)I.

Abstract

HeLa cell 5 S ribosomal RNA, labeled in vitro at 40°, 60°, or 70°C with (125)I, has been digested with RNase T1 and fingerprinted by conventional methods. Regardless of the temperature of iodination, (125)I was bound (exclusively) to virtually all CMP residues. Characterization of labeled oligonucleotides yielded sequence information consistent with that derived from in vivo labeled RNA.
