Percentage of the dilution recovery of the P5CP assay.
| Undiluted | 100% | 100% | 100% | 100% |
| Dilution 1:2 | 107 | 98 | 95 | 93 |
| Dilution 1:4 | 122 | 109 | 118 | 89 |
| Mean | 109 | 102 | 104 | 94 |
Notes: Measurements refer to duplicates. Undiluted sample represents 100% recovery, from which recovery is calculated for the remaining dilution steps from 1:2 up to 1:4.
Abbreviations: HS, human serum; RS, rat serum.
Inter- and intra-assay variation for the P5CP assays using human serum as quality control samples.
| HS1 | 4.9 | 6 | 11.1 |
| HS2 | 4.2 | 2.6 | 9.4 |
| HS3 | 5.8 | 3.2 | 7.6 |
| HS4 | 6.5 | 4.2 | 9.1 |
| HS5 | 7.4 | 3.5 | 7 |
| HS6 | 3.7 | 8.2 | 9.6 |
| HS7 | 4.9 | 5.9 | 11.2 |
| HS8 | 5 | 5.7 | 7.7 |
| Mean | 4.9 | 9 |
Note: The variation was calculated as the mean variation of 10 individual determinations of each sample.
Figure 1Serum P5CP levels during progression phase at 4 (n = 12), 6 (n = 13) and 8 (n = 20) weeks of CCl4 treatment.
Notes: Regression phase was monitored in animals that were treated with CCl4 for 8 weeks after which they were left to relapse for 14 (n = 7), 20 (n = 17) or 26 (n = 18) weeks.
ROC values of the P5CP marker during progression and regression phases.
| Control vs. combined 4–8 weeks | 0.85 | 0.05 | <0.0001 | 91.1 | 52.3 | |
| Control vs. CCl4 4 weeks | 0.83 | 0.09 | <0.05 | 83.3 | 57.1 | |
| Control vs. CCl4 6 weeks | 0.93 | 0.05 | <0.001 | 92.3 | 42.8 | |
| Control vs. CCl4 8 weeks | 0.82 | 0.1 | <0.05 | 95 | 57.1 | |
| CCl4 vs. combined 14–26 weeks | 0.92 | 0.03 | <0.0001 | 96 | 60 | |
| CCl4 vs. 14 weeks regression | 0.80 | 0.10 | <0.05 | 85.7 | 60 | |
| Control vs. 20 weeks regression | 0.95 | 0.03 | <0.0001 | 94.2 | 60 | |
| Control vs. 26 weeks regression | 0.97 | 0.02 | <0.0001 | 100 | 60 |
Notes: Cut off values of 5.4 ng/mL and 9.2 ng/mL was decided for the progression and regression phase respectively, and the representing values of sensitivity and specificity are presented.
Figure 3Total collagen deposition in liver slices as seen with Sirius red staining. rats which received no CCl4 showed no visible collagen deposits or fibrotic band formation (A). Animals receiving a weekly dosage of CCl4 for 4, 6 or 8 weeks showed increased collagen deposition and fibrotic band formation (B). rats which were treated for 8 weeks with CCl4 after which they were left to regress for 14, 20 or 26 weeks, revealed a gradual decrease in the total collagen deposition and fibrotic band formation (C).
