Abstract
This essay chronicles the major theoretical and experimental contributions made by Charles A. Figure 2Top panel: Note that lone engagement of co-stimulatory molecules (signal 2) has no effect on a T cell. Bottom panel: On the other hand, recognition of antigen (signal 1) in the absence of co-stimulatory molecules — as might occur in vivo when autoreactive T cells bind to self antigens — results in “anergy,” a permanent state of unresponiveness which disables the T cell’s ability to mount an immune attack. Anergy is one mechanism by which autoimmunity is avoided.
