Abstract
The upstream stimulatory factor 1 (USF1) gene has been shown to play an essential role as the cause of familial combined hyperlipidemia, and there are several association studies on the relationship between USF1 and metabolic disorders. In this study, we analyzed two single nucleotide polymorphisms in USF1 rs2073653 (306A>G) and rs2516840 (1748C>T) between the case (dyslipidemia or obesity) group and the control group in premenopausal females, postmenopausal females, and males among 275 Korean subjects. We observed a statistically significant difference in the GC haplotype between body mass index (BMI) > or =25 kg/m2) and BMI <25 kg/m2 groups in premenopausal females ( chi2=4.23, p=0.04). It seems that the USF1 GC haplotype is associated with BMI in premenopausal Korean females.Entities:

Clinical characteristics of study subjects
Data were shown as mean±standard deviation (SD). Analysis of variance (ANOVA) test was done among 3 groups. *, p<0.05; †, p<0.01; ‡, between premenopausal and postmenopausal female; §, between premenopausal female and male; ‖, between postmenopausal female and male.
BMI, body mass index; BP, blood pressure; LDL, low-density lipoprotein; HDL, high-density lipoprotein.
Frequencies of genotype and haplotype, and linkage disequilibrium in the subjects
Haplotypes were constructed with rs2073653+rs2516840.
Comparison of phenotypes among genotypes in three groups
Data were shown as mean±standard deviation (SD). Analysis of variance (ANOVA) test was done among 3 genotypes (GG, GA, and AA) of rs2073653, and Student t-test was done between 2 genotypes (CC and CT) of rs2516840 in each three group.
BMI, body mass index; LDL, low-density lipoprotein; HDL, high-density lipoprotein; TG, triglyceride.
Statistical analysis results of frequencies of genotype and allele between case and control groups of each phenotype in three groups
p*, p value (df=2); p†, p value (df=1). OR, the ratio of odds for the minor allele of each SNP (A allele in rs2073653 and T allele in rs2516840) in the contro group to the odds for the major allele. CI, confidence interval; BMI, body mass index; LDL, low-density lipoprotein; HDL, high-density lipoprotein.
Haplotype analysis and association with BMI in premenopausal female
Haplotypes were constructed with rs2073653+rs2516840; *Likelihood ratio test with each haplotype and the others to compare the case (BMI ≥25 kg/m2) group and the control (BMI <25 kg/m2) (df=1).
BMI, body mass index.
Comparison of haplotype frequencies between case and control groups of each phenotype in three groups
Haplotypes were constructed with rs2073653+rs2516840; *Likelihood ratio test with each haplotype and the others to compare the case and the control (df=1). BMI, body mass index; TG, Triglyceride; LDL, low density lipoprotein; HDL, high density lipoprotein; p, p value.
