Figure 1Our preliminary study found that Swiss 3T3 fibroblasts displayed good reproducibility in both the production and the suppression of ROS, by paraquat and by antioxidants respectively. NAC at concentrations of 10~40 mM suppressed ROS in a dose.dependent manner, while 50% suppression was achieved by 10 mM NAC.
Figure 2Effect of Cys, Cys2, and Met on the intensity of ROS in Swiss 3T3 fibroblasts. Intensity of ROS was measured by a laser scanning confocal microscope. Y axis is intensity values folds of control value. Cys, Cys2, and Met were observed to suppress ROS at various concentrations in a dose dependent manner: both Cys and Met at 1~1000 µM, and Cys2 at 40~400 µM. 50% suppression was achieved by 10 µM of Cys and Met, and by 400 µM of Cys2.
Mean concentration of amino acids and glutathione (µM /L) following intravenous administration of 25 mg/kg of NAC
SD, standard deviation; *cystine concentrations were lower than basal level (p<0.05).
Figure 3Plasma concentration of reduced NAC and total NAC following intravenous administration of 25 mg/kg in seven healthy volunteers. The t1/2 was 2.88±1.14 h for NAC and 3.68±1.84 h for total NAC. The CL was 1.23±0.77 L. h-1 kg-1 for NAC and 0.56±0.27 L. h-1 kg-1. for total NAC. The Vss was 3.07±0.10 L kg-1. for NAC and 3.00±0.11 L kg-1. for total NAC. Total NAC is the sum of the reduced NAC and oxidized NAC.
